Abstract
Event Abstract Back to Event Combatting the impact of aging on recovery from brain injury Jolien M. Van Houcke1, Eve Seuntjens2 and Lut Arckens1* 1 KU Leuven, Biology, Belgium 2 KU Leuven, Biology, Belgium Traumatic brain injury (TBI) has a large impact on victim and society and will be one of the main causes of death and disability by 2020, yet effective treatment is unavailable. Its increased prevalence in elderly, with a worsened outcome, urges the use of a model in which the cellular and molecular components of neuronal regeneration can be unravelled in an aging context. We use the extraordinary short-lived African Turquoise killifish (N. furzeri), a rapid aging model and thus ideal to unravel the impact of aging on successful regeneration upon injury. By introducing a stab-wound in the telencephalon of young and aged fish, we provide a fast and robust TBI model to elucidate the underlying cellular and molecular processes of de novo neurogenesis, differentiation, outgrowth and synaptogenesis, and how they are affected with age. We will explore the contribution of radial glia and peculiar, abundant highly-proliferative neuronal progenitor cells (hpNPCs) in neuroregeneration of the telencephalon. More importantly, the molecular players that differentiate the injury repair between young and aged fish will be identified via a functional proteomics approach targeted towards identifying drugable plasma membrane proteins, and validated via pharmacological interventions or genetic manipulation in our optimized TBI model.
