Abstract
Temporomandibular joint osteoarthritis (TMJOA) is characterized by progressive osteochondral degeneration, and the efficacy of current intra-articular clinical treatments remains inconsistent. Fisetin (FST) has shown therapeutic promise in osteoarthritis. In this study, an monosodium iodoacetate (MIA)-induced TMJOA rat model was used to evaluate the reparative effects of intra-articularly administered FST on condylar cartilage damage. The results demonstrated that FST exhibited a transient improvement in TMJOA-associated abnormalities in cartilage thickness, but the effect did not persist long-term. To address the pharmacokinetic limitations of FST, FST-loaded poly(lactic-co-glycolic acid) nanoparticles (FST-PNP) were prepared using a single emulsion solvent evaporation method. The nanoparticles exhibited uniform morphology and favorable sustained-release properties, which markedly extended the release duration of FST. These results indicate that a sustained-release of FST from poly(lactic-co-glycolic acid) (PLGA) nanoparticles can be a promising material for TMJOA treatment.
